COMT and psychiatric disorders
The COMT Val158Met polymorphism, resulting in the substitution of valine (Val) for methionine (Met) at position 158 of the COMT enzyme, is associated with differences in dopamine levels and neurotransmitter function. Individuals with the Val/Val genotype, associated with higher COMT enzyme activity, may be at increased risk for psychiatric disorders.
Slow COMT (met/met):
- better performance on
attention-related tasks
- improved working memory
- more sensitive to stressful
events, higher cortisol (children, adolescents)
- more likely to avoid harm (less
novelty seeking)
- increased relative risk of OCD,
panic disorder, pain disorders
Fast COMT (val/val):
- less cognitive decline in aging
- less sensitive to stressful
events
- better at processing aversive
emotional stimuli
- greater extroversion and more
novelty seeking
- increased relative risk of
ADHD, substance abuse
